In vitro and in vivo evaluation of the antiangiogenic activities of Trigonella foenum-graecum extracts

Objective To assess the antiangiogenic activity of fenugreek. Methods Different fractions of fenugreek crude extracts were prepared and their antiangiogenic properties were assessed using the ex vivo rat aortic ring assay and in vivo chicken embryo chorioallantoic membrane (CAM) assay. They were investigated for their direct cytotoxic activity in the MCF7 cells using the MTT assay. Results The ethanol extract showed 100% inhibition of blood vessel outgrowth from primary tissue explants in the rat aortic ring assay at a concentration of 100 μg/mL while the other extracts did not show significant antiangiogenic activity. The ethanol extract was therefore investigated at varying concentrations and exhibited a significant dose dependent effect. The CAM assay coincided with the results of the aortic ring assay as ethanol extract showed a significant inhibition of formation of new blood vessels. The extracts only showed anti-proliferative activity at the highest concentration of 400 μg/mL towards MCF7 breast cancer cell lines in the MTT assay. Conclusions Findings of the both assays confirmed that the ethanol extract inhibited vascularization significantly. Further studies on the ethanol extract would be beneficial in isolating the active ingredient responsible for the inhibition.

16S rDNA-based metagenomic analysis of human oral plaque microbiota in patients with atherosclerosis and healthy controls.

To address the question if an altered oral microbiota is associated with atherosclerosis. Twenty patients suffering from atherosclerosis and 10 controls were recruited. Clinical oral, medical and laboratory investigations were performed. Oral bacteria were collected and 16S rDNA was sequenced following Single strand conformation polymorphism.(SSCP) Probing pocket depths in patients were significantly elevated. The oral microbiota of patients and controls were dominated by Fusobacterium (16%/17%), Streptococcus (21%/14%), Prevotella (10%/12%), Enterococcus (12%/12%), Porphyromonas (8%/7%), TM7 (0%/7%) and Veillonella (6%/7%). Differences in diversity were not significant between groups. The pathology of atherosclerosis may not be related to significant qualitative changes of the oral microbiota.

Radiologia da prótese total do quadril e suas complicaçöes / Imaging of total hip arthroplasty and related complications

Resumo: A utilizaçäo da prótese total do quadril (PTQ) em pacientes coma anormalidades da articulaçäo coxofemoral é um procedimento bastante difundido. No entanto, apresenta uma série de complicaçöes, como desprendimento, infecçäo, ossificaçäo heterotópica, luxaçöes e fraturas, que säo facilmente detectadas por radiografias simples e outros métodos de imagem. Neste artigo säo descritos a técnica radiográfica ideal para avaliaçäo da PTQ, os achados radiológicos normais no pós-operatório indicam a presença de complicaçöes. O papel de métodos complementares, como cintilografia óssea e a artrografia, também é analisado.

Sarcoma granulocítico(cloroma)-relato de um caso / Gramulocytic sarcoma (chloroma)-a case report

Resumo: O sarcoma granulocítico, conhecido também como cloroma, é um tumor sólido raro, composto de células imaturas da série granulocítica, que ocorre usualmente como complicaçäo de leucemia mielóide aguda. Mais raramente astá associada a leucemia mielóide crônica e outros disturbios mieloproliferativos. Apresentamos um caso de sarcoma granulocítico multifocal de manisfestaçäo pouco comum, tanto pelos órgäos envolvidos como também por manifestar-se sem associaçäo com quadro leucêmico.
